Real-time polymerase chain reaction for identification of dog meat in adulterated beef meatball using specific primer targeting on cytochrome-b for halal authentication

This research aimed to validate real-time polymerase chain reaction using specific primer targeting on cytochrome-b 33 (cyt-b 33). The primer cyt-b 33 could specifically amplify dog meat DNA and method could detect 0.5 pg dog meat DNA in meatball formulation. The repeatability assay was acceptable with RSD value ≤ 25%. The validated method was further used for detection of meatball products commercially sold in Yogyakarta Indonesia, and results revealed that all samples (n = 14) did not contain dog meat DNA. This research finding supports the implementation of Indonesian Law Number 33 of 2014 in developing method for Halal Product Assurance.
