A Single Bout of Exercise Inhibits Alpha-2-HS-Glycoprotein (fetuin A) in the Adipose Tissue of Obese Mice

PURPOSE In a recent study, alpha-2-HS-glycoprotein (fetuin A) was reported to disrupt the energy homeostasis of adipose tissue. Therefore, we tried to determine if a single bout of exercise could inhibit fetuin A in adipose tissue. In addition, we investigated the correlation between plasma concentration and expression in adipose tissue of fetuin A. METHODS Male C57BL/6 mice were divided into four groups; normal diet (ND) control (n=7), ND exercise (n=7), high-fat diet (HFD) control (n=7), and HFD exercise (n=7) groups. All exercise mice ran on a treadmill at 15 m/min for 60 minutes for single bout. All animals were sacrificed two hours after single-bout of exercise and adipose tissue was isolated. RESULTS Fetuin A expression in adipose tissue was significantly lower in HFD-EX than in HFD control (p<.001). Concentrations of fetuin A in plasma were lower in HFD-EX than in HFD control (p<.001). However, correlation analysis showed that there was no correlation between protein expression in adipose tissue and plasma concentration in the other groups except to HFD-EX. CONCLUSIONS A single bout of exercise effectively inhibited fetuin A in adipose tissue of obese mice. Therefore, a single bout of exercise seems to regulate energy homeostatic defects in adipose tissue through fetuin A normally. In addition, there was no absolute correlation between the expression of fetuin A in adipose tissue and the plasma concentration, so cannot represent each other.
